Important notes on CO2 cylinder usage:
Never use CO2 cylinder outside of working hours. Make sure the CO2 sensor is on all the time you are using the cylinder. In the case of a gas leak (the sensor alarm is activated): Turn off the gas cylinder valve (if possible) and leave the room immediately. During office hours, contact SCELSE Lab Management at 6592 3029 / 6592 7560. After office hours, contact NTU Campus Security at 6790 5200.

Setting up CO2 cylinder:

• Bring cylinder from Nitrogen Gas Yard to ABIF and chain it with the trolley to a table leg.
• Connect the tube attached to the cylinder to the “100% CO₂ IN" push-in connector on the tube attached to Tokai Hit main unit.
• Ensure CO₂ gas sensor is present and turned on.
• Turn gas cylinder valve anticlockwise (open to maximum) to release gas to the system.
• The gas regulator valve has been preset to 1 bar. There is no need to adjust the valve. If the right-hand side pressure gauge shows more than 1 bar, contact ABIF staff or Lab management immediately.

Setting up Tokai Hit:

• If using CO₂, connect the soft tubing attached to “MIX GAS OUT” port of the main unit to the stage-top incubator chamber.
  
• Turn on the power switch of the main unit (1).
• If using an immersion lens, attach the lens heater (velcro band) to the objective lens and turn it on (2).
• If you want to use the sample temperature as a feedback for the heater, place the thermocoupler (a thin wire) inside the stage-top chamber and switch the feedback control on (3). Do not switch it on if the thermocoupler is not inside the chamber or else the room temperature will be used as feedback and the heater will heat until the maximum temperature possible before giving an error.
• If using CO₂, turn on the “MIX GAS POWER” switch (4).
• Set the desired temperature and CO2 concentrations using the buttons next to the respective displays on the main unit.

Shutting down Tokai Hit:

• Turn off the “MIX GAS POWER” switch (4).
• Turn off the power switch of the main unit (1).

Removal of CO₂ cylinder:

• Turn the gas cylinder valve clockwise completely to close the gas supply.
• Remove the tube from the push-in connector.
• Push the blue ring towards the black part and it will be possible too pull out the tube without much force.
• A short gush of CO₂ will come out when tubing is removed. If CO₂ is continually leaking, tighten the gas cylinder valve again.
• Unchain the gas cylinder and bring it back to Nitrogen Gas Yard. Do not leave the gas cylinder in ABIF overnight.
